SNAP-cell products contain cell-permeable sustrates for rapid precision labeling of SNAP-tag fusion proteins inside live cells. It is ideal for protein localization experiments, protein translocation, flow cytometry studies, and more. In addition, certain SNAP-vitro substrates, which are not cell-permeable, are particularly suited to exclusively label extracellular domains of membrane proteins. This labeling approach can be used for example to follow receptor internalization. SNAP-tag labeling has worked well in all mammalian cell lines tested so far including CHO-K1, COS-7, HEK 293, HeLa, NIH 3T3, V79, and U-2 OS. The SNAP-tag is based on the DNA repair protein AGT, but because it is highly optimized for reaction with SNAP-tag substrates, no background labeling from wtAGT is seen ( further information).
